If I registered for Qantas’ triple points promotion and then book the flights through the Amex travel portal using my $450 credit from the Qantas Ultimate credit card, would that booking be eligible for the triple points?

Flights can be booked via qantas.com, a travel agent or Qantas Contact Centres.
So I think that’s fine to book via Amex Travel, as they are acting as a travel agent.
